Diagnostic Biosystems High Contrast DAB (Liquid Format)
3,3'-Diaminobenzidine (DAB), is a widely used chromogen for immunoperoxidase staining, and is well accepted among pathologists because of its increased sensitivity and decreased background when compared to amino ethylcarbazole (AEC). Specimens stained with DAB can be dehydrated, cleared, and permanently mounted. High Contrast DAB is more sensitive and stable than traditional working DAB solutions. High Contrast DAB offers several noteworthy improvements and benefits as compared to traditional working DAB solutions. High Contrast DAB is highly sensitive, providing the cost-effective option of further diluting the primary antibody. Being stable for 2 weeks (as opposed to 6 hours for traditional DAB working solutions), High Contrast DAB allows the user the convenience of making one working solution for the entire week’s work. Hazardous waste generation from unused DAB solution is also significantly reduced.

Podoplanin identifies the ~38 kDa O-linked transmembrane sialoglycoprotein podoplanin, which is expressed in the endothelium of lymphatic capillaries, but not in the blood vasculature (1). Besides the expression in lymphatic endothelium, podoplanin is also found in a variety of other tissues, including mesothelial cells, reticular cells, follicular dendritic cells, ovarian and testicular germ cells (2). Results aid in the classification of lymphatic invasion of primary tumors.

MSH6 is a heterodimer of MSH2 and binds to DNA containing G/T mismatches. The MSH2-MSH6 complex recognizes a single-base mispair insertion/deletion loop. An alteration of microsatellite repeats is the result of slippage owing to strand misalignment during DNA replication and is referred to as microsatellite instability (MSI). These defects in DNA repair pathways have been related to human carcinogenesis. Studies have shown the mutations of MLH-1, MSH2 and MSH6 genes contribute to the development of sporadic colorectal carcinoma. The repair of mismatch DNA is essential to maintaining the integrity of genetic information over time.

Smooth muscle myosin heavy chain (SM-MHC) is a cytoplasmic structural protein which is a major component of the contractile apparatus in smooth muscle cells. Expression of smooth muscle myosin is developmentally regulated, appearing early in smooth muscle development, and is specific for smooth muscle development. Two isoforms of smooth muscle myosin heavy chain have been identified, designated MHC-1 and MHC-2, they are differentially expressed depending on the functional activity of the muscle.\nThe antibody may be useful for the study of breast tumors as the presence of an intact layer of myoepithelial cells is an important feature, which may distinguish benign breast lesions and carcinoma in situ from invasive tumors
Diagnostic Biosystems Solution Pepsin
Pepsin is used for proteolytic digestion of formalin-fixed paraffin-embedded (FFPE) tissue sections prior to application of antibodies. In immunohistochemistry (IHC), most commonly used fixatives such as formalin mask tissue antigens (cellular, membrane, and nuclear) by their intrinsic crosslinking. This masking results in poor or no staining in IHC. Pepsin digestion of FFPE tissue sections improves accessibility of antibodies to tissue antigens.\n\nFixation is one of the most critical aspects of immunostaining. If the antigen is not properly fixed it will be washed out of the specimen. On the other hand, overfixation can cause severe problems such as masking or denaturation of the antigen. Formaldehyde is probably the best all-around fixative, due to its cross-linking characteristics. Length of fixation is very critical to prevent antigen masking.
